ABSTRACT

Objective To investigate the combined application value of tandem mass spectrometry ( MS/MS) and high performance liquid chromatogram-tandem mass spectrometry ( LC-MS/MS ) in the newborn screening of children with methylmalonic acidemia ( MMA ) . Methods The dried blood spot samples from the newborn with abnormal propionylcarnitine ( C3) or C3/acetylcarnitine ( C2) or C3/free carnitine ( C0) levels in the preliminary screening of MS/MS were collected, and the concentrations of MMA, methylcitric acid ( MCA) and homocysteine ( Hcy) in these samples were detected with LC-MS/MS. The neonates with increased MMA, MCA or Hcy levels were recalled, and their urinary organic acids were analyzed with gas chromatographic mass spectrometry (GC/MS). Last, gene mutation anal-ysis was performed to make a definite diagnosis.Results A total of 423 samples with abnormal C3 or C3/C2 or C3/C0 levels in the new-born screening were collected, and the positive rate of preliminary screening was about 1%. The LC-MS/MS results showed that 8 neo-nates had higher MMA and tHcy levels. The GC/MS results further showed that the level of MMA increased slightly. Conclusion The combined application of MS/MS and LC-MS/MS may increase the positive predictive value and decrease the false positive rate of MMA screening in the newborn, which may have an important clinical significane in the screening of inherited metabolic disorders.

ABSTRACT

OBJECTIVE: To determine the concentrationsof antofloxacin hydrochloride in the lung tissuesafter single dose preoperative oral administration of 400 mg in patientsreceiving pulmonry lobectomy for, preliminary evaluation of the rationality of this regimen to prevent infection. METHODS: Forty-eight patients with no clinical evidence of preoperative infection undergoing pulmonary lobectomy were selected and randomized into four treatment groups. 400 mg antofloxacin hydrochloride was administered orally at 4, 8, 12, and 24 h before the removal of the bronchi. Blood samples and lung tissues were collected after operation. The antofloxacin hydrochloride in blood serum and lung tissues was determined by high performance liquid chromatogram. RESULTS: The average concentrations of antofloxacin hydrochloride in serum andlung tissuesin the four groupswere (3.281±0.817)μg·mL-1 and (5.992±3.228)μg·g-1, (3.192±0.953)μg·mL-1 and (5.885 ±2.952)μg·g-1, (1.712 ±0.576) μg·mL-1 and (3.419±1.107)μg·g-1, and (0.431±0.095)μg·mL-1 and (1.258±0.306) μg·g-1, respectively. CONCLUSION: The clearance time of oral antofloxacin hydrochloride in lung tissue was longer than that in serum. Single dose administration of oral antofloxacin hydrochloride 400mg within 12 h before operation can achieve effective bactericidal levels in serum and lung tissues, which could be a method to prevent infection for pulmonary lobectomy.

ABSTRACT

Objectives To investigate the characteristics of changes in serum metabolic profile before and after resection of carcinoma tissues to establish a disease distinguishing model,to analyze the changing trend of characteristic metabolites,and to determine the molecular mechanism and potential clinical value of characteristic metabolic markers for HBV-related liver cancer.Methods Ultraperformance liquid chromatography-mass spectrometry (UPLC-MS) was used to analyze the serum metabolites of 15 patients with hepatocellular carcinoma (HCC) before and after partial hepatectomy and on 25 healthy volunteers.The pattern recognition method and nonparametric test analyzes were used to analyze the data and to identify the specific metabolites and their changes after resection of carcinoma tissues.Results We established the principal component analysis (PCA) and orthogonal partial least squares-discriminant analysis (OPLS-DA) disease distinguishing model for HCC patients before and after operation as against the healthy volunteers.To distinguish between the liver cancer group and the normal control group,27 characteristic metabolites were selected from the patients before and after resection of carcinoma tissues.Eight moved towards the normal control after resection of carcinoma tissues.This indicated that liver carcinoma was an important impacting factor for these metabolites.Finally,7 metabolites were identified,and these metabolites had high diagnostic value as shown on ROC curves.Conclusions Through serum metabolic profiling of patients before and after resection of carcinoma tissues,a high correlation between metabolism and hepatocellular carcinoma was found.Researches on endogenous metabolites and pathways in liver diseases will provide a better understanding of the molecular mechanisms and provide further directions for clinical diagnosis and treatment.

ABSTRACT

Objective: To study the purification of Sini Decoction (SND) with macroporous resin D 101 . Methods: SND was isolated and purified by macroporous resin D 101 . The purification of SND was evaluated by chemical and pharmacological methods. Results: The thin layer chromatograms, high performance liquid chromatograms of the SND purification and those of SND were almost the same; also the SND purification can significantly promote the content of nitric oxide (NO) in serum of mice treated by Metaraminol. Conclusion: The SND purification can keep the active ingredients of SND.

ABSTRACT

Objective To establish a method of high performance liquid chromatogram(HPLC) for measuring matrine and oxymatrine in Zhidaipian Tablets.Methods A Lichrospher-NH2 column(4.6 mm?250 mm,5?m) was used with a mobile phase consisting of acetonitrile-ethanol-0.5 mol/L phosphoric acid(80∶10∶10),and in a flow rate of 1.0 mL/min.The detective wavelength was 212 nm.Column temperature was room temperature and the injection quantity was 20 ?L.Results The samples were well separated.The linear range was the same for the target(1.0-10.0 ?g/mL).The linear equations for matrine was C=1.201?10-4A +0.161,the correlation coefficient was 0.9992,and for oxymatrine was C=1.366?10-4A +0.221,while the correlation coefficient was 0.9996,respectively.The mean recovery rate were 99.9% and 99.4%,respectively.RSD was 1.48% and 4.33%,respectively(n=6).Conclusion The method is simple,reliable,accurate and can be used to the quality control of the Zhidaipian Tablets.
